Celladhesionmoleculesoftenplayanessentialroleincell-to-cellinterac-
tions.Twolympho-hematopoieticcellscanonlyestablishcontactifoneof
themexpressessurfacemoleculesthatinteractwithligandsexpressedon
thesurfaceoftheothercell.AsforAPCandTcellinteractions,theresult
ofsuchcontactmaybethatasignalcapableofinducingdifferentiation
andfunctionalchangeswillbeinduced.Adhesionproteinsareusuallycom-
prisedofseveralchainswhichcaninducedifferenteffectswhenpresentin
variouscombinations.Interactionofseveralcascadesisoftenrequiredfor
thefinaldifferentiationofacell.Celladhesionmoleculesnormallyform
partoftheIgsuperfamily(e.g.,ICAM,VCAM,CD2),integrinfamily(lympho-
cytefunctionantigen,LFA-1),selectinfamily,cadherinfamily,orvarious
otherfamilies.Selectinsandintegrinsalsoplayanimportantroleininter-
actionsbetweenleukocytesandthevascularwall,andthusmediatethemi-
grationofleukocytesfromthebloodstreamintoinflamedtissues,ortheentry
ofrecirculatinglymphocytesintothelymphnodeparenchymathroughhigh
endothelialvenules(HEV).
Chemokines(chemoattractantcytokines)compriseafamilyofover 30 small
(8– 12 kDa)secretedproteins.Thesecontributetotherecruitmentof“inflam-
matorycells”(e.g.,monocytes)intoinflamedtissues,andinfluencetherecir-
culationofallclassesofleukocytes(Table2. 6 ).Somechemokinesresultinthe
activationoftheirtargetcellinadditiontoexertingchemotaticproperties.
ChemokinescanbeclassifiedintothreefamiliesbasedontheirNterminus
structure:CCchemokinesfeaturetwocontiguouscysteineresiduesattheter-
minus;CXCchemokineshaveanaminoacidbetweenthetworesidues;and
CX3CandCchemokinesthusfarcompriseonlyonemembereach(fractalkine
andlymphotactin,respectively).AlthoughtheNterminuscarriesbioactive
determinants,usingachemokinesaminoacidsequencetopredictitsbiolo-
gicalfunctionisnotreliable.Thechemokinesystemformsaredundantnet-
work,orinotherwords,asinglechemokinecanoftenactuponanumberof
receptors,andthesamereceptormayrecognizeanumberofdifferentche-
mokines.Manyofthechemokinesalsooverlapintermsofbiologicalfunction.
Chemokinescanbegroupedintwofunctionalclasses:inflammatoryche-
mokineswhicharesecretedbyinflamedorinfectedtissuesasmediatorsof
thenonspecificimmuneresponse;andconstitutivechemokineswhichare
producedinprimaryorsecondarylymphoidorgans.Togetherwithendothe-
lialadhesionmolecules,inflammatorychemokinesdeterminethecellular
compositionoftheimmigratinginfiltrate.Incontrast,thefunctionofconsti-
tutivechemokinesistodirectlymphocytestopreciselocationswithinlym-
phoidcompartments.Thus,chemokinesplayamajorroleintheestablish-
mentofinflammatoryandlymphoidmicroenvironments.Chemokinerecep-
torsareGprotein-coupledmembranereceptorswithseventransmembrane
sequences.Inkeepingwiththeabovenomenclature,theyaredesignatedas
CCR,CXCR,orCX3CRplusconsecutivenumbering.Someviruses,forinstance
