C. diphtheriaeproduces black colonies.
Promotes development of metachromatic granules.
Corynebacteriumspp produce gray to black colonies due to
reduction of tellurite. C. diphtheriaecolonies are surrounded
by a brown halo.
Bismuth sulfite & brilliant green inhibit most others. S. typhi
colonies are black, surrounded by metallic sheen. Others are
light green. (Some salmonellae may be inhibited.)
Crystal violet inhibits most GN. Novobiocin inhibits GPC.
Cefsulodin inhibits most GP & GN. Y. enterocoliticaferments
mannitol, appears as red “bull’s-eye” colonies surrounded by
colorless halo.
Alkaline pH suppresses commensals. Subcultured to TCBS.
