period with the stain. In order to avoid high background
staining, additional washes may be performed as described in
steps 11 and 12. For staining vacuoles through the stress-
recovery time course described in Subheading 3.1 , incubate
the cells with the stain for a minimum of 1 h before the 42 °C
for 1 h treatment.
