  1. BMDM medium: DMEM containing 4.5 g/L glucose and
    1 mM sodium pyruvate, 10% (v/v) heat-inactivated FBS,
    100 units/mL penicillin, and 100μg/mL streptomycin.

  2. L929-conditioned medium.

  3. Hemocytometer.

  4. Trypan blue.

  5. Benchtop microcentrifuge.

  6. 2 cm cell scraper.


2.2 Cholesterol

Incorporation)



  1. Cells of interest: BMDM.

  2. ClassІІbiological safety cabinet (BSCІІ).

  3. Bead bath (hot water bath) set at 37C.

  4. Humidified 5% CO 2 incubator at 37C.

  5. 12-Well tissue culture plates.

  6. BMDM medium.

  7. A-769662: 50 mM A-769662 in dimethyl sulfoxide (DMSO)
    (or AMPK activator of choice,seeTable1).

  8. Sodium acetate (500 mM).
    9.^3 H-Sodium acetate (75–150 mCi/mmol) (Perkin Elmer).

  9. DMSO.

  10. Sterile 1PBS.

  11. Sterile 1.5 mL microfuge tube.

  12. 1 cm cell scraper.

  13. Methanol/chloroform (2:1) (v/v).

  14. Chloroform.

  15. Double-distilled H 2 O (ddH 2 O).

  16. Thin-layer chromatography (TLC) plates (silica gel 60).

  17. Solvent system: heptane/isopropyl ether/acetic acid (60:40:3)
    (v/v/v).

  18. Cholesterol standard.

  19. Hamilton syringe.

  20. Nitrogen gas.

  21. Iodine crystals.

  22. Flat-edged razor blade.

  23. Weigh paper.

  24. Scintillation cocktail.

  25. 7 mL scintillation vials (or equivalent).

  26. Scintillation counter (e.g., Tri-Carb 2910 TR, Perkin Elmer).

  27. Bicinchoninic acid assay (BCA) protein determination kit
    (or equivalent assay).
