- Plasma samples from multiple collection tubes can be pooled
for increased DNA yields. - In order to prevent contamination by leukocytes or other
DNA containing cells, it may be useful to leave a small layer of
untouched plasma behind at the plasma/cell interface.
Pipetting should be performed slowly to avoid disturbing the
cell layer and operators may consider drawing up the first ~75%
of available plasma first before attempting the last ~25%. - Use of liquid nitrogen freezing and longer centrifugation in
the final steps will lead to increased DNA yield, but also higher
proportions of contaminants. Adjust accordingly.
