- The Personal Genome Machine (PGM) (Fig. 4a) should be
maintained including weekly chloride washes when the
machine is in use and water washes after each run. - Before each run, the PGM should be initialized in strict accor-
dance using sequencing reagents kit. In addition, the pressure
of gas tank should be checked (Fig. 4b). - Before each run, a run plan should be generated using the Ion
Torrent browser on the server.
3.3 Running
of Sequencer

it measures the hydrogen ions produced during DNA replication. The upper compartment (arrow) is where the
chip is placed. The four ports shown represent the four deoxynucleotide (dNTP) (A, T, C, G) ports where the
correct reagent bottle representing the nucleotides should be attached. (b) Nitrogen gas cylinder (arrow) is a
requirement for the working of benchtop sequencer. The gas tank should be changed when the pressure is
below 500 psi; it is important for the initialization of the sequencer. (c) Chip for sequencing. It consists of load-
ing port (arrow) where the pipette is inserted firmly for the delivery of ion sphere particles for sequencing on
the sequencer. (d) Grounding plate on the benchtop sequencer. Chip (Fig. 5 ) has to be placed on the grounding
plate (arrow) in order to avoid damage to the chip due to electrostatic discharge
