Procedure is the same as explained for ALP activity except the use of citrate buffer
instead of carbonate-bicarbonate buffer.
27.6 Calculations
The enzyme activity is calculated in terms of KAU as explained for ALP. Plot a
graph between OD and the amount of phenol. Activity can also be calculated using
equation explained for ALP.
27.7 Clinical Significance...............................
Normal range of ACP is 1–3.5 KAU/dl. ACP levels increase 40–50 times of normal
levels in prostate cancer. Earlier, ACP was used for diagnosis of prostate cancer that
is now replaced by prostate-specific antigen. Slight increase in ACP activity is also
observed in hyperparathyroidism with skeletal involvement, malignant inversion of
bones by cancers such as female breast cancer, and Paget’s disease. Non-prostrate
ACP fraction increases in myelocyte leukemia and some other hematological
disorders.
