Thereisnosurfacemarkerwhichcanunequivocallydifferentiatebetween
memoryTandBcellsand“naive”(neverbeforeactivated)cells.Instead,im-
munologicalmemoryisnormallytakentocorrelatewithanincreasednum-
berofspecificprecursorTandBcells.Followinganinitialimmunizationwith
antigen,thisincreasedprecursorfrequencyofspecificcellsisthoughttobe
maintainedbyanantigen-independentprocess.Yettheprecursorcellscan
onlybeactivated(orre-activated)byantigen,andonlyactivatedTcellscan
provideimmediateprotectionagainstre-infectionoutsidethelymphoidor-
gans,e.g.,inthesolidperipheralorgans.Similarly,onlyantigenactivated
Bcellscanmaturetobecomeplasmacellswhichmaintaintheincreasedblood
antibodytitersresponsibleformediatingprotection.Thisindicatesthatre-
sidualantigenmustbepresenttomaintainprotectiveimmunologicalmem-
ory.Asageneralrule,thelevelofprotectiveimmunitymediatedbytheex-
istenceofmemoryTandBcellsperseisminimal.Highlyeffectiveimmunity
andresistancetore-infectionareinsteadprovidedbymigratoryTcellswhich
havebeenrecentlyactivated(orre-activated)byantigen,andbyantibody-
secretingBcells.B-cellandantibodymemoryismaintainedbyre-encoun-
terswithantigen,orbyantigen-IgGcomplexeswhichbyvirtueoftheir
FcportionsorbybindingtoC3barecapturedby-,andmaintainedfor
longperiodson-,folliculardendriticcellspresentingerminalcenters.Mem-
oryTcells,andinsomecasesBcells,canbere-stimulatedandmaintained
inanactivestateby:persistentinfections(e.g.,tuberculosis,hepatitisB,HIV);
antigendepositsinadjuvants;periodicantigenre-exposure;peptide-loaded
MHCmoleculeswithlonghalf-lives;orpossibly(butrarely)bycross-reactive
antigens.Thus,secondarilyactivated(protective)memoryTandBcellscan-
noteasilybedistinguishedfromprimarilyactivatedTandBcells.Theanti-
gen-dependentnatureofimmunologicalprotectionindeedquestionsthe
relevanceofaspecialized“memoryquality”ofBandTcells.

ItisimportanttodifferentiatebetweenthecharacteristicsofmemoryTand
Bcellsasdetectedinvitro,andthesalientin-vivoattributesofimproved
immunedefenses.Followingaprimaryimmuneresponse,increasednum-
bersofmemoryBcellscanofcoursebedetectedusinginvitroassaysor
bymurineexperimentsinvolvingthetransferofcellsintonaiverecipients.
However,theseincreasedBcellfrequenciesdonotnecessarilyensureim-
muneprotectionagainst,forinstance,viralre-infection.Suchprotection
requirestheexistenceofanincreasedtiterofprotectiveantibodieswithin
thehost.
