  1. Ramp rate for the entire cycle is set to 2 °C/s to ensure even
    heat distribution throughout the droplets.
    REMEMBER! Set the sample volume to 40 μL (this is an
    approximate value for the total volume of droplets produced
    from the emulsion of the generator oil and reaction mixture)
    and the heated lid to 105 °C.

  2. The lowest most cluster of droplets are the nontarget contain-
    ing negative droplets, while the next two bands of increased
    fluorescence amplitude were comprised of droplets that pos-
    sessed only one target type. The final population of droplets
    contain both target sequences and consequently had the great-
    est fluorescence signal.

  3. Note that primer concentration range written is just as a sug-
    gested guideline and further optimization may be required by
    increasing the range. However, as a starting point, the sug-
    gested primer concentration range may be used.
