Drug Metabolism in Drug Design and Development Basic Concepts and Practice

(nextflipdebug2) #1

18.4.2.2 Urine The HPLC radiochromatographic profiles of pooled urine
from rats, dogs, monkeys, and humans were also determined (Table 18.7).

18.4.2.3 Feces and Bile Excretion in feces represented more than 95% of
the recovered radioactive dose in rats, dogs, monkeys, and humans.
Muraglitazar was one of the abundant components present in feces from all
species. The HPLC-radiochromatograms of pooled bile samples from rats,
monkeys, and humans are shown in Fig. 18.3. The relative distribution of
metabolites in urine and feces (as % of dose) is shown in Table 18.7.

TABLE 18.7 Relative distribution (percent of dose) of radioactive metabolites in
pooled urine and feces after an oral dose of [^14 C]muraglitazar to intact rats, dogs,
monkeys, and humans.

aMetabolites are assigned based on retention times of radioactive peaks and LC/MS analysis.
bRadioactive peaks are reported as a percentage of dose (% of the sample multiplied by % excretion).
cTrace¼<0.01% radioactivity and detected by LC/MS.
dND¼not detected. Criteria are no discernable radioactive peak,<0.01% radioactivity and not
detected by LC/MS.
eUncharacterized peaks.
