AMPK Methods and Protocols

(Rick Simeone) #1

  1. DAPI is a cell permeable fluorescent probe for DNA. It allows
    nuclear labeling.

  2. Once adenoviral construct is used, several biosafety precautions
should be taken. All work with adenovirus minimally requires
BSL-2 containment. People must wear protective equipment
to reduce mucosal or skin exposure. Working surfaces must be
disinfected after use. Wastes must be disinfected by bleaching
    and/or autoclave.

  3. The optimal temperature for luciferase activity measurement is
    room temperature.

  4. ERK1/2 phosphorylation is acutely and only temporarily sti-
mulated by pro-hypertrophic stimulation. This stimulation
tempts to disappear after longer incubation time such as 24 h.
